LEADERSHIP IN FIBROSIS: Promedior's New Therapeutic Paradigm

Armed with unique and comprehensive insights into monocyte biology, Promedior is creating a therapeutic platform based on pentraxin-2 (PTX-2) , a naturally-occurring protein shown to regulate a number of important immune, inflammatory, and fibrotic processes in the body. Promedior drug developers have discovered a natural pentraxin-based mechanism through which the body regulates the fundamental innate immune response at the source by controlling monocyte and monocyte-derived cell differentiation and activation mechanisms. Promedior’s therapeutics use this natural mechanism to specifically recognize a site of injury and control how the monocytes react to damaged tissue, stimulating monocyte differentiation into a beneficial, pro-resolution regulatory state and preventing differentiation into pathologic fibrocytes and M2 macrophages.

PTX-2 Naturally Targets Monocytes

PTX-2 Naturally Targets Monocytes: Pentraxin-2 (PTX-2) simultaneously controls the initiation and progression of fibrosis and promotes healing and resolution. In the presence of PTX-2, recruited monocytes are signaled by the PTX-2 coated cellular debris to differentiate into regulatory macrophages, locally shifting the cellular equilibrium to resolution and healing. The regulatory macrophages secrete IL-10 to inhibit myofibroblast-driven scar tissue production and shift the ratio of TIMP/MMP expression resulting in increased break down of scar tissue. MMP, matrix metalloproteinase; TIMP, tissue inhibitor of MMP.

Fibrosis is the common pathway in virtually all forms of chronic organ failure, including kidney, liver, lung and heart failure. Fibrotic organ failure affects more than 20 million people in the US alone.
